La Ligne directrice décrit l'essai in vivo de mutation du gène Pig-a sur érythrocytes de mammifères qui utilise un gène endogène de mammifère, le gène de classe A phosphatidylinositol-glycane (Pig-a), comme rapporteur de mutation d'un gène de cellule somatique. Les tests de mutation génique in vivo tels que l'essai Pig a se prêtent particulièrement bien à l'évaluation de la mutagénicité, car une série de facteurs physiologiques tels que l'absorption du produit chimique d'essai à partir du site d'exposition, sa distribution dans l'ensemble du système d'essai par la circulation systémique, ainsi que les processus in vivo du métabolisme et de la réparation de l'ADN, contribuent aux réponses mutagéniques. L'essai Pig-a peut être pratiqué sur les souches communément utilisées de rats ou de souris (1) (voir paragraphe 28), et ne nécessite pas d'euthanasier les animaux. Ces caractéristiques facilitent l'intégration de l'essai Pig a à de nombreux protocoles d'essai in vivo sur des rongeurs.

This Test Guideline proposes defined approaches (DA) combining data generated in vitro methods, with information sources such as physicochemical properties. The prediction from a DA may be used alone to determine eye hazard potential according to the hazard classes of the UN GHS (Categories 1, 2, or not classified). A DA consists of a fixed data interpretation procedure (DIP) (i.e. a mathematical model, a rule-based approach) applied to data (e.g in silico predictions, in chemico, in vitro data) generated with a defined set of information sources to derive a prediction without the need for expert judgment. The DAs use method combinations intended to overcome some of the limitations of the individual, stand-alone methods in order to provide increased confidence in the overall obtained result.
